Clarke was born in India in 1899. He joined the British Army in 1918 and served in the Royal Engineers. After the war, he worked as a journalist and businessman. In 1939, he was recalled to active duty and joined the Intelligence Corps.

Clarke's first major deception operation was Operation Fortitude, which was designed to deceive the Germans about the location of the Allied invasion of Normandy. Clarke created a number of fake army units and equipment and deployed them in southeastern England, making it appear that the Allies were planning to invade France there. The Germans were fooled by the deception and concentrated their forces in Normandy, which allowed the Allies to land successfully.

Clarke's most famous deception operation was Operation Mincemeat, which was carried out in 1943. In this operation, Clarke planted a dead body on a beach in Spain with fake documents that suggested that the Allies were planning to invade Greece. The Germans found the body and the documents, and they believed the deception. This helped to convince the Germans that the Allies were not planning to invade Italy, which allowed the Allies to land successfully in Sicily.
